ChIP-seq and RNA-seq profiling of mice hippocampus before and after Arid1a conditional knockout.

To gain a deeper insight into roles of Arid1a in mice hippocampus development, we performed ChIP-seq and RNA-seq to analyze the genome-wide changes of histone marks and transcriptome changes before and after Arid1a deletion in hippocampus of the E16.5 and P21 mouse using Cre-lox system. Total RNA was extracted from WT and Arid1a-cKO hippocampus and generated by deep sequencing, in two or three biological replicates, using BGISEQ-500. ChIP-seq was sequenced without biological replicates on BGISEQ-500 platform .

为深入阐明Arid1a在小鼠海马发育中的功能机制,我们借助Cre-lox系统,在胚胎第16.5天(E16.5)及出生后第21天(P21)的小鼠海马组织中特异性敲除Arid1a,随后通过染色质免疫共沉淀测序(ChIP-seq)与RNA测序(RNA-seq)技术,全基因组层面分析该基因缺失前后的组蛋白修饰标记变化与转录组表达谱改变。我们从野生型(WT)与Arid1a条件性敲除(Arid1a-cKO)小鼠的海马组织中提取总RNA,采用BGISEQ-500平台进行深度测序,设置2~3个生物学重复。ChIP-seq实验未设置生物学重复,同样在BGISEQ-500平台完成测序。
