Juvenile hormones (JH) and ecdysteroids regulate many biological and metabolic processes. CREB-binding protein (CBP) is involved in co-activation of many transcription factors in different organisms. We studied the role of CBP in JH action in the red flour beetle, Tribolium castaneum. CBP knockdown caused a decrease in JH induction of genes, Kr-h1, 4EBP and G13402 in T. castaneum cell lines. Trichostatin A [TSA, a histone deacytelase (HDAC) inhibitor] induced the expression of these JH response genes. Furthermore, RNA seq and subsequent differential gene expression analysis revealed the impact of CBP RNAi on the global gene expression in Tc cell lines. The CBP RNAi affected the expression of genes coding for transcription factors and other proteins associated with growth and development when comparing with malE RNAi cells. CBP RNAi has also shown negative impact on JHIII and TSA induction of genes. These data suggest a role of CBP in JH action and development in the model insect, T. castaneum.
