U87-MG wild-type (WT) and ∆α cells were treated with 150 µM H₂O₂ for 2 hours. Five days post-treatment, the cells were fixed and stained according to the manufacturer’s instructions using the Senescence Cells Histochemical Staining Kit (C030, Sigma-Aldrich). This kit detects senescent cells based on β-galactosidase activity at pH 6, which is present in senescent, but not in quiescent, immortal, or tumor cells. Cellular senescence is characterized by irreversible growth arrest, morphological changes, and altered gene expression patterns. Images of stained cells were captured using an Olympus microscope.
