Gene expression profiling reveals a potential role of Tieghemelin and Maslinic acid in stimulating hair growth in C3H mice. C3H male mice are a model for hair growth, treated with 1% Tieghemelin, 1% Maslinic acid and 1% Minoxidil for 20 days. Microarray gene expression profiling was conducted for two biological replicates Collected mice skin samples were subjected to DNA microarray analysis. Template RNA was extracted using ISOGEN solution (Nippon Gene, Japan) as described above. The GeneChip WT PLUS Reagent Kit (ThermoFisher Scientific, Waltham, MA, USA) and GeneChip™ Hybridization, Wash and Stain Kit (ThermoFisher Scientific, Waltham, MA, USA) were utilized following the manufacturer's protocol to generate a purified, fragmented, and labeled aRNA. The Clariom S array Mouse (ThermoFisher Scientific, Waltham, MA, USA) was used for hybridization in the GeneChip™ Fluidics Station (ThermoFisher Scientific, Waltham, MA, USA). The arrays were scanned using the GeneChip Scanner (ThermoFisher Scientific, Waltham, MA, USA), generating raw signal intensity values (CEL files). The CEL files were normalized using the Transcriptome Analysis Console (TAC) Software (version 4.0.1). Genes with fold change ≥ 1.7 (in linear space) and p-value ≤ 0.05 were considered differentially expressed genes (DEGs) and were used for further analysis.
