RNASeq was performed on Huh-7 human hepatocarcinoma cells to determine basal gene expression levels. The data were then used to filter the results of a genome-wide siRNA screening for lipoprotein uptake.

A high-throughput genome-wirde siRNA screening for lipoprotein uptake was performed on Huh-7 human hepatocarcinoma cells. In order to exclude possible false-positives coming from siRNA molecules targeting genes which are actually not expressed in Huh-7 cells, we determined basal gene expression in this cells line by RNASeq on an Illumina platform. Five samples were sequenced with increasing read depth in order to ensure that all low-expressed genes were detected. Expression levels from the sample with the highest read depth were used to filter the siRNA screening data.
