Effective FTO inhibitors with potent anti-tumor efficacy suppress cancer stem/initiating cell self-renewal and overcome immune evasion

We conducted RNA sequencing with poly A RNA isolated from NOMO-1 cells upon shRNA induced FTO knockdown and small molecules (CS1 and CS2) mediated FTO inhibition . Overall design: The NOMO-1 cells were infected with lentivirus targeting FTO and the stable cells were selected with 1ug/ml puromycin for 1 week. As FTO inhibitors treatment, the NOMO-1 cells were treated with 100nM CS1 and 500nM CS2 for 72 hours. Then, the total RNAs were extrated from each samples and polyA RNAs were enriched for sequencing. Each group contains at least 3 biological replicates.
