Differential miroRNA expression in round spermatids of GRTH null mice compared to wild type

miRNA expression profiles for round spermatids of wild type and GRTH knock-out mice were determined by Rodent TaqMan® Low Density miRNA Arrays A v2.0 (TLDA, Applied Biosystem). Purified round spermatids were prepared from the testis of wild type and GRTH null mice (C57BL/6 strain). Equal amount of total RNA from 20 mice (wild type or GRTH KO) was pooled prior to gene expression analysis.本研究采用啮齿类TaqMan®低密度miRNA芯片A v2.0（Rodent TaqMan® Low Density miRNA Arrays A v2.0，简称TLDA，应用生物系统公司（Applied Biosystem）），对野生型（wild type）与GRTH基因敲除（GRTH knock-out）小鼠的圆形精子细胞（round spermatids）的miRNA表达谱进行检测。研究人员从C57BL/6品系（C57BL/6 strain）野生型及GRTH基因缺失（GRTH null）小鼠的睾丸中分离纯化得到圆形精子细胞。将20只小鼠（野生型组或GRTH KO组）的等量总RNA（total RNA）混合后，开展基因表达分析（gene expression analysis）。
