Transcriptomic analysis of injured and uninjured knee joints from Trem2 KO mice [bulk RNA-seq]

Trem2 is a single-pass transmembrane receptor of the immunoglobulin superfamily and is primarily expressed in myeloid cells. Cells from injured and uninjured knee joints of wildtype C57BL/6 mice and Trem2 KO mice were sequenced to determine if Trem2 plays a role in macrophage differentiation or function after joint injury. Right knee joints of 10-week-old C57BL/6 and Trem2 KO mice were injured using a tibial compression injury model. Uninjured and 7-day post-injury joints were collected and cells were analyzed using bulk RNA-seq.
Trem2是免疫球蛋白超家族(immunoglobulin superfamily)的单次跨膜受体(single-pass transmembrane receptor),主要在髓系细胞中表达。为探究Trem2在关节损伤后是否参与巨噬细胞分化或功能调控,研究人员对野生型C57BL/6小鼠及Trem2敲除(knockout, KO)小鼠的受损与未受损膝关节细胞进行了测序。实验中,研究人员采用胫骨压迫损伤模型,对10周龄的C57BL/6及Trem2 KO小鼠的右膝关节造模,随后分别收集未损伤状态及损伤后7天的膝关节组织,并通过批量RNA测序(bulk RNA-seq)对其中的细胞开展分析。
