We evaluated promoter behavior in a pooled context. All 11 promoter constructs were detected in the bulk population by short-read sequencing. Cells were then binned into four mStayGold-based expression clusters using flow cytometry (negative, low, medium, and high), and the specific promoter sequences in each sorted cluster were investigated by next generation sequencing.
