Our study reports that transient delivery of BCL-XL increases survival of human induced pluripotent stem cells (iPSCs) by ~10-fold after plasmid transfection, leading to a 20- to 100-fold increase in HDR (homology directed repair) mediated knockin efficiency and a 5-fold increase in knockout efficiency. We examined the transcriptomes of survived iPSCs at 2 hour, 4 hour and 8 hour after electroporation with or without BCL-XL overexpression by RNA-seq.
