Purpose: The goal of this study is to profile genes expression in mice liver during liver regeneration process post 2/3 partial hepatectomy using RNA-seq. Methods: Mice aged 8~10 weeks were performed with either 70% partial hepatectomy(PH) or shamed operation. At 0.5h, 2h, 4h, 24h, 48h, 72h, 120h, 168h post PH and after shamed operation, mice (six at each time point) were killed and liver samples were harvested. Total RNA were prepared from the liver samples using TRIZOL reagent (Invitrogen), and the mRNA profiles were genenated by Illumina HiSeq 2000. qRTâPCR validation was performed using TaqMan and SYBR Green assays. Results: Our study provides a realiable dataset covering the three phases of liver regeneration: primer phase(0.5h,2h,4h), proliferation phase (24h, 48h, 72h), and termination phase (120h,168h). Using this dataset, we discover some interesting facts undiscovered before, especially on the re-organization process after proliferation wave and the termination phase. Overall design: mRNA profiles at 0.5h, 2h, 4h, 24h, 48h, 72h, 120h, 168h post hepatectomy and sham group were generated by deep sequencing, each group has two pooled samples (three samples were pooled into one).
