We provide evidence highlighting a novel mechanism by which choline remodels CRC and upregulate the SE-driven key transcription factor KLF5 via the methyl donor pathway, implicating potential therapeutic targets in HCC. Whole-transcriptome sequencing (RNA-Seq) was perfemed to identify the downstream target genes regulated by choline in Huh7 cells. Chromatin immunoprecipitation followed by sequencing (ChIP-seq) was performed using indicated antibodies to study the H3K27ac and H3K4me1 status and binding sites of transcription factor in Huh 7 cells under 0μM and 15μM choline treatment. Cleavage Under Targets and Tagmentation (CUT&Tag) was performed to explore whether the super-enhancers or promoters within downstream target genes are directly regulated by KLF5.
