记录冰川、径流、土壤及空气微生物样品的原始采集过程。1)冰雪微生物样品采集:采集过程中佩戴洁净手套,将冰雪采集至洁净自封袋中。2)冰尘微生物样品采集:通过软管插入冰尘穴底部,使用注射器将底泥、融水吸入到采样瓶中,低温保存带回实验室,底泥和冰尘穴上部融水均用来提取环境DNA。3)径流包括冰上径流和冰川前缘径流,直接采集径流融水至采样瓶或者采水袋中。4)冰川前缘土壤的采集:使用铲子采集土壤样品,通过2mm土筛后将土壤装至洁净的whirl-pak采样袋中,然后低温保存,用于后续土壤DNA的提取。5)空气膜样品采集:将设计好的采样装置置于采样点,装置下部为电池(可持续工作48h),上部包含两张滤膜收集空气微生物,用于DNA的提取。6)冰川径流、融水中理化性质的实时监测,使用YSI多参数水质仪,直接放入待测样品中,获取温度、DO、叶绿素浓度等。
Documents the original sampling procedures for microbial samples collected from glaciers, runoff, soils and air. 1. Collection of snow/ice microbial samples: Wear sterile gloves during sampling, and transfer the collected snow and ice into sterile self-sealing bags. 2. Collection of cryoconite microbial samples: Insert a hose into the bottom of the cryoconite hole, aspirate the sediment and meltwater into the sampling bottle using a syringe, store the samples at low temperature and transport back to the laboratory. Both the sediment and the upper meltwater in the cryoconite hole are used for environmental DNA extraction. 3. Runoff includes supraglacial runoff and glacial frontal runoff: Collect the runoff meltwater directly into sampling bottles or water collection bags. 4. Collection of glacial frontal soil samples: Use a shovel to collect soil samples, pass the samples through a 2mm soil sieve, then transfer the treated soil into sterile Whirl-Pak sampling bags, and store at low temperature for subsequent soil DNA extraction. 5. Collection of air filter membrane samples: Place the pre-designed sampling device at the sampling site. The lower part of the device is equipped with a battery that can operate continuously for 48 hours, while the upper part contains two filter membranes for collecting air microorganisms, which are used for DNA extraction. 6. Real-time monitoring of physico-chemical properties in glacial runoff and meltwater: Use a YSI multi-parameter water quality meter, immerse the instrument directly into the sample to be tested, and obtain parameters such as temperature, DO, and chlorophyll concentration.
