We injected adeno-associated viruses (AAV2/5) carrying an inhibitory peptide derived from ß-adrenergic receptor kinase 1 (ißARK), tagged with mCherry for identification, into the medial prefrontal cortex (mPFC) of 5xFAD mice. This strategy aimed to attenuate Gq GPCR-induced Ca2+ activity in astrocytes. In the control group, mice were injected with AAV2/5 expressing only mCherry. Subsequently, snRNA sequencing analysis was conducted on the mPFC tissue samples from these mice, enabling a comparative assessment of transcriptomic alterations and an exploration of the potential implications resulting from the modulation of Gq-GPCR signaling. Overall design: Comparative gene expression profiling analysis of snRNA-seq data for WT-mCherry, WT-ißARK, 5xFAD-mCherry, and 5xFAD-ißARK.
