To examine the potential off-target effects by CRISPR/Cas13d-mediated HoxB13 knockdown in 293FT cells. To analyze differentially expressed genes upon CRISPR/Cas13d-mediated HoxB13 knockdown in LNCaP95 mouse liver metastasis. 293FT cells were transfected with non-targeting pre-gRNA control(gCtrl) or pre-gRNA targeting HoxB13(gHoxB13) expression plasmid in combination with Cas13d expression plasmid for 24h. LNCaP95 mouse liver metastasis samples were collected after mice treated with LNP-Cas13d-pre-gControl or SCORT LNP-Cas13d-pre-gHoxB13 for 72h in the final treatment.RNA-seq was performed to characterize the differentially expressed genes between gCtrl and gHoxB13 groups.
