The Supplementary Data related to the paper State-specific extraction of environmental DNA: Spike-and-recovery controls to validate and optimise extraction protocols, Environmental Microbiology 2025, contain the following: Sheet 1: Percent recovery of exDNA and iDNA spike-ins in both target and nontarget eDNA states depending on the spiked eDNA state, the bacterial origin, the selected extraction protocol as well as the environmental samples tested. Esco_iDNA and Basu_iDNA refer to the iDNA spike-ins originating from E. coli and B. subtilis, respectively. Similarly, Esco_exDNA and Basu_exDNA represent the spiked exDNA deriving from each of the model organisms. Sheet 2: Total percent recovery of exDNA and iDNA spike-ins irrespective of their presence in the target and nontarget eDNA state. Values are listed separately for each of the spiked eDNA states, their bacterial origins, the selected extraction protocols (including also the extraction of the total eDNA using a commercial DNA extraction kit) as well as the environmental samples tested. Esco_iDNA and Basu_iDNA refer to the iDNA spike-ins originating from E. coli and B. subtilis, respectively. Similarly, Esco_exDNA and Basu_exDNA represent the spiked exDNA deriving from each of the model organisms. Sheet 3: Physicochemical and microbiological properties of the selected environmental samples. For DHA (dehydrogenase activity), Na+, NH4+, K+, Mg2+, Cl-, NO2-, NO3-, PO43-, SO42-, values refer to the fresh weight (FW) of samples.
