Mature optic nerve regeneration requires ligand-independent expression of the LIF receptor

To investigate the molecular consequences of LIFR expression, RGCs were FACS-isolated from vglut2-tdTomato mice two weeks following intraocular AAV2-LIFR, AAV2-Ctrl or AAV2-shLIFR, AAV2-shScramble and then analyzed by bulk RNA-seq. AAV2-LIFR and AAV2-shLIFR had a pronounced transcriptional effect on adult RGCs compared to the control group. To compare transcriptional changes in adult RGCs isolated from mice two weeks following intravitreal injection of AAV2-LIFR, AAV2-Ctrl, AAV2-shLIFR or AAV2-shScramble (AAV2-LIFR vs. AAV2-Ctrl; AAV2-shLIFR vs. AAV2-shScramble, n=3 replicates each group).
