This dataset contains the raw and processed data underlying the manuscript “Humanized yeast to study the role of human ECE-1 isoforms in apoptosis in congenital heart disease” by Hanhui Xie et al. The study establishes a humanized Saccharomyces cerevisiae model co-expressing human Bax, Bcl-xL and ECE-1 isoforms to investigate mitochondrial apoptosis and outer mitochondrial membrane homeostasis in a genetically tractable system. File 1 – Figures and Tables (submission version) This file provides the final versions of Figures 1–13 and Tables 1–2 used in the journal submission, together with the corresponding figure legends. The figures include: colony PCR verification of diploid genotypes (Fig. 1), spot assays validating Bax/Bcl-xL toxicity and rescue (Fig. 2), standard curve and ACT1 control for semi-quantitative PCR (Fig. 3), expression profiles of apoptotic regulators YCA1, NUC1 and BI-1 (Fig. 4), TOM, SAM and TIM complex subunits under Bax/Bcl-xL expression (Figs. 5–7), ECE-1 isoform effects on Bax-induced growth inhibition by spot assay and growth curves (Figs. 8–9), and expression changes in apoptotic regulators and mitochondrial translocase components under Bax+ECE-1c conditions (Figs. 10–13). Tables 1 and 2 list all primers used for plasmid construction, colony PCR and semi-quantitative PCR analysis. File 2 – Original uncropped agarose gels (Supplementary file) This file contains the full-length, uncropped agarose gel images corresponding to the semi-quantitative PCR and colony PCR experiments shown in Figures 1, 3–7 and 10–13. For each gel, all edges are visible and lanes are annotated with figure numbers, gene names and treatment conditions where applicable. Additional unused biological replicates with similar banding patterns are provided to illustrate the reproducibility of the assays. These images document the underlying data for grayscale quantification of PCR products (measured in ImageJ) and subsequent statistical analyses (one-way ANOVA performed in GraphPad Prism 7). The dataset is intended to support transparency and reproducibility of the Bax/Bcl-xL/ECE-1 yeast system and the associated analyses of mitochondrial apoptotic regulators and protein-import machineries (TOM, SAM and TIM complexes). Users are welcome to re-use these data for re-analysis, method comparison or meta-analyses, provided that the original article and this dataset are appropriately cited.
