Sequencing of histone H3.3-HaloTag incorporated genomic regions.

Recent advanced live-cell imaging techniques have revealed the dynamic behavior of chromatin in living cells. Especially, our single-nucleosome imaging studies have revealed the genome-wide local chromatin motion and its heterogeneity, which can provide a higher-order regulation of genome functions. Individual nucleosomes in living cells are commonly labeled by tagged core histones (e.g., histone H3.3 tagged with HaloTag). To determine the genomic localization of the nucleosomes labeled with H3.3-HaloTag in HeLa cells, we pull-downed the H3.3-HaloTag nucleosomes and sequenced their genomic DNAs.
