We report the application of single-molecule-based sequencing technology for high-throughput profiling of mouse pulmonary arteries (main, right and left extralobar branches) from C57BL/6 mice exposed to 6 weeks or hypoxia (FiO2 10%) and normoxia (FiO2%) Overall design: adult male mice (C57BL/6J; 14 weeks old, Jackson Laboratory, Bar Harbor, ME) were housed in an antigen-free and virus-free animal care facility under a 12-hour light and dark cycle. Two mice were exposed to hypoxic conditions by placing them in a plexiglass chamber connected to OxyCycler Models A420OC and AT42CO gas controllers and analyzers (BioSpherix, NY, USA). This gas control delivery system regulated the flow of room air, N2, and O2 into the chamber to create and maintain prescribed fraction inspired O2 (FiO2) levels. Persistent hypoxia defined by a FiO2 of 10% was maintained for 6 weeks at 24 hours/day. mice were fed a standard rodent chow and had free access to water. Animals were euthanized with overdose of urethane by intraperitoneal injection followed by exsanguination, followed by harvest of the pulmonary arteries.
