The role of TEFM in mitochondrial gene expression [TruSeq]

We investigated the role of TEFM in vivo by creating heart- and skeletal-muscle specific knockout of this gene. We identified that TEFM is required for the processivity of POLRMT and its loss leads to significantly decreased mitochondrial transcription. Total RNA from isolated heart mitochondria from four control mice (L/L) and four Tefm knockout mice (L/L, cre), was sequenced and analysed for differential expression.本研究通过构建该基因的心脏与骨骼肌特异性敲除模型，在活体水平探究了TEFM的生物学功能。研究证实，TEFM是POLRMT持续合成能力所必需的，其缺失会导致线粒体转录水平显著降低。我们对4只对照小鼠（L/L）与4只Tefm敲除小鼠（L/L, cre）分离得到的心脏线粒体中提取的总RNA进行了测序，并开展差异表达分析。
