To overexpress TaCSN5-2A in wheat, the coding sequence was amplified and inserted into the pWMB110 vector driven by the maize ubiquitin promoter. For RNAi induced gene silencing, a hairpin RNA silencing fragment was designed and inserted into the pWMB110 vector. The recombinant constructs were transformed into immature embryos of the wheat variety Fielder by Agrobacterium mediated transformation.Fielder and transgenic TaCSN5 plants were cultured in Hoagland solution for 7 days with 16 hours light and 8 hours dark photoperiod at 23 degree. Wheat seedlings were treated with 250 mM NaCl for 48 hours and then sampled for RNA seq analysis. Total RNA was isolated using RNA Plus reagentaccording to the manufacturers instructions. Three biological replicates were assayed. The RNA-seq was conducted on the BGISEQ-500 platform.
