Effects of Salidroside on gene expression in ischemic brain of rats

We employed DNA microarray to identify genes differentially regulated by salidroside in ischemic brain of MCAO rats. With a cut of p < .01 and 2-fold change, we found that 121 genes were upregulated and 142 genes were down-regulated in the MCAO group, compared to the sham group. Administration of salidroside (1 h after reperfusion, daily for 6 d) enhanced 28 transcripts and suppressed 16 transcripts. The genes involved in neuroplasticity and oxygen carrier were further confirmed by qRT-PCR. Our data suggest that salidroside could regulate neuroplasticity in post-ischemic stroke, in addition to its neuroportective function.

本研究采用DNA微阵列(DNA microarray)技术,筛选大脑中动脉闭塞(MCAO, Middle Cerebral Artery Occlusion)大鼠缺血脑组织中受红景天苷(salidroside)调控的差异表达基因。以p<0.01且表达变化倍数≥2作为筛选阈值,与假手术组相比,MCAO模型组中共鉴定出121个上调表达基因及142个下调表达基因。于再灌注后1小时开始给予红景天苷,每日给药1次,连续给药6日,该给药方案可上调28个转录本的表达,同时下调16个转录本的表达。针对参与神经可塑性与氧载体功能的基因,本研究通过qRT-PCR(quantitative real-time polymerase chain reaction)进行了进一步验证。本研究结果表明,红景天苷除具备神经保护功能外,还可调控缺血性脑卒中后的神经可塑性过程。
