Amplicon sequencing of 16S rRNA and PsbA gene, prokaryote and phytoplankton assemblages in sediments and seawater near Nagatsura-Ura Lagoon, Japan

The aim of this study is to use NGS to analyze the horizontal distribution and behavior of the bacterial community in sediments and the phytoplankton assemblages in seawater during autumnal anoxia or hypoxia in the inner part of the lagoon in comparison with periods when there is no anoxia or hypoxia. Sediment and seawater samples were collected in Nagatsura-Ura Lagoon. After DNA was extracted from all samples, the 16SrRNA and PsbA gene were amplified with primers (341F-805R, and PsbAyo1F-PsbAyo1R) using PCR. The PCR product mixtures were analyzed for pair-end DNA sequencing of 300 bp Ã? 2 using MiSeq desktop sequencer (Illumina). The sequenced data were analyzed in the Qiime bioinformatics pipeline.
