RNA-seq analysis of sorted CD45+CD11b+Ly6G+ cells from C57BL/6 and Traj18-/- CRC samples and healthy controls

We performed RNA-seq analysis of tumor infiltrating neutrophils from C57BL/6 mice and iNKT deficient Traj18-/- mice to understand the role of iNKT cells in affecting phenotype and functions of neutrophils in CRC. We observed that TANs from B6 animals were enriched for transcripts of chemokines and inflammation as well as of immune suppression. These data suggest that iNKT cells condition the phenotype of TANs.本研究对C57BL/6小鼠及恒定自然杀伤T细胞（invariant natural killer T cells, iNKT）缺陷型Traj18-/-小鼠的肿瘤浸润中性粒细胞（tumor infiltrating neutrophils, TANs）开展RNA测序（RNA-seq）分析，旨在明确恒定自然杀伤T细胞在结直肠癌（colorectal cancer, CRC）中对中性粒细胞表型与功能的调控作用。本研究观察到，B6小鼠的肿瘤浸润中性粒细胞中，趋化因子、炎症相关及免疫抑制相关的转录本均呈现富集状态。上述实验数据表明，恒定自然杀伤T细胞可调控肿瘤浸润中性粒细胞的表型。
