Impact of the Gram-negative-selective inhibitor MAC13243 on the in vitro simulated gut microbiota

We investigated the impact of the LolA inhibitor MAC13243 on the human gut microbiota by using an in vitro model simulating colon passage (CoMiniGut). We characterised the effects of this compound on microbiota composition and diversity by performing viable cell counts, 16S rRNA gene quantification and sequencing, and MIC testing of E. coli isolates. The results show that exposure of the gut microbiota to MAC13243 inhibits E. coli growth without significantly reducing microbial diversity and relative abundance of beneficial taxa, which are all favourable traits for preventing antibiotic-associated dysbiosis.
