Muscle glucocorticoid receptor (GR) signaling regulates gene expressions in skeletal muscle. To reveal the roles of muscle GR in systemic metabolism in obesity, ob/ob background muscle-specific GR knockout (ob/ob GRmKO) mice were generated. GR Flox+/+ Cre-/- Lep+/+ (lean GRf/f), GR Flox+/+ Cre+/- Lep+/+ (lean GRmKO), GR Flox+/+ Cre-/- Lepob/ob (ob/ob GRf/f), and GR Flox+/+ Cre+/- Lepob/ob (ob/ob GRmKO) were compared. Gastrocnemius muscle from 7-week-old male mice was excised and analyzed. We used pooled RNA samples (n = 3).
肌肉糖皮质激素受体(glucocorticoid receptor, GR)信号通路调控骨骼肌的基因表达。为阐明肥胖状态下肌肉GR在全身代谢中的作用,本研究构建了ob/ob背景的肌肉特异性GR敲除(muscle-specific GR knockout, GRmKO)小鼠模型。实验对以下四组小鼠进行比较:GR Flox+/+ Cre-/- Lep+/+(瘦型GRf/f)、GR Flox+/+ Cre+/- Lep+/+(瘦型GRmKO)、GR Flox+/+ Cre-/- Lepob/ob(ob/ob型GRf/f)以及GR Flox+/+ Cre+/- Lepob/ob(ob/ob型GRmKO)。采集7周龄雄性小鼠的腓肠肌(gastrocnemius muscle)组织并进行分析,本研究采用混合RNA样本(n=3)。
