The goals of this study are to understand how the chikungunya virus (CHIKV) RNA-dependent RNA polymerase, nsP4, regulates genome replication and fidelity. We generated nsP4 variants, transfected these in BHK-21 and C6/36, and 48 hours later harvest the supernatant for deep sequencing of the CHIKV genome. These studies are important to define how the polymerase functions to maintain fidelity during infection.
