Expression data from Tbx1 knockout (KO) palatal shlelves

Deletion of Tbx1, a member of the T-box transcription factor gene family, results in abnormal epithelial fusion between the palatal shelves and the mandible, which induces cleft palate by inhibiting elevation of the palatal shelves. We used microarrays to determine the downstream genes of Tbx1 during palatogenesis and identified distinct classes of dysregulated genes. Wild-type and Tbx1KO head or palatal shelves were collected at Embryonic day (E) 12.5 or 13.5 for RNA extraction and hybridization on Affymetrix Mouse Gene 2.0ST Array.
作为T盒(T-box)转录因子基因家族成员的Tbx1基因缺失,可导致腭突与下颌骨之间出现异常上皮融合,并通过抑制腭突抬升过程诱发腭裂。本研究采用基因芯片技术,筛选腭发育过程中Tbx1的下游靶基因,鉴定出多类表达失调的基因。我们于胚胎第12.5天(E12.5)或第13.5天(E13.5)收集野生型与Tbx1敲除(Tbx1KO)小鼠的头部组织或腭突,用于RNA提取,并在Affymetrix小鼠基因2.0ST阵列(Affymetrix Mouse Gene 2.0ST Array)上完成杂交实验。
