We aimed to investigate both dosing time and allelic-dependent variations in a widely used anticancer drug oxaliplatin pharmacokinetics and drug-induced toxicity, and identify the mechanistic basis by RNA sequencing. Cryptochrome double knockout (Cry1-/-Cry2-/- DKO) or wild-type (C57BL/6J, WT) mice kept in constant darkness period (DD) for three days and were treated with a single dose oxaliplatin (12 mg/kg, i.p.) at two different circadian times (CT8 and CT16). Drug induced toxicity was assessed by mortality and body weight loss. Oxaliplatin pharmacokinetics was evaluated for each group. RNA sequencing was performed in the liver of mice following 12 mg/kg i.p. oxaliplatin injections for three consecutive days.
