IIn contrast to the young mice treated with anti-PD-1 therapy, aged mice exhibited immune-related adverse event (irAE)-like symptoms in the lung when treated with anti-PD-1 therapy. To understand the molecular events underlying the development of anti-PD-1 therapy-induced irAE, we employed deep RNA sequencing of whole lung transcript from anti-PD-1 therapy-treated young and aged mice. In order to further evaluate the effect of IL-21 signal on PD-1-blockade-mediated immune response, gene expression profile in lung tissue from aged mice twith the blockade of IL-21 activity was also assessed. Whole lung transcripts were obtained from melanoma-bearing young or aged mice treated anti-PD-1 Ab, aged mice treated with rat control Ab, and aged mice similtaneously treated with anti-PD-1 Ab and anti-IL-21 receptor in vivo. Each gene expression profile was analyzed by RNA sequencing, and the differential gene expressions in whole lung tissues among those stimulatory conditions were assessed.
