In this work we propose a multicomponent MSC-B cell coculture system to expand and study in vitro MSC induced Breg (iBreg). Tonsillar B cells were stimulated with anti-IgM, a CD40 agonist and IL-2 for 7 days. We further asked whether comparative RNAseq study of sorted IL-10+/- iBreg identify novel Breg biomarkers.
