16S rRNA V3-V4 amplicon sequencing of Holstein dairy cows rumen microbiota

The effect of ginkgo fruits on Holstein dairy cows rumen microbiota was evaluated by the rumen simulation technique (RUSITEC). Rumen content was collected from 2 ruminally fistulated Holstein cows and strained through 2 layers of surgical gauze. The rumen inoculum was a mixture of rumen fluid and artificial saliva at a 5:2 ratio. Continuous culture by RUSITEC was performed using a rumen inoculum containing 1.6% of ginkgo extract and one without adding anything. Samples were collected on the last 2 days of culture and used for microbial analysis. Culture was performed in quadruplicate.
