Human engineered CRC organoids were equiped with the sTomato-p27K- quiescense reporter. Quiescent cells are characterized by distinct transcriptional programs Overall design: Organoids were grown from single cells for 7-10 days. Then, organoids were dissociated to single cells and sTomato-p27k-high, sTomato-p27k-medium, and sTomato-p27k-low cells were collected by FACS. Samples were then processed for bulk RNA-seq.
