Single-cell gene expression profiling of CD4+ T cells from the murine small intestine lamina propria [steady-state_SI-Treg_10XscRNAseq]

Intestinal regulatory T cells consist of at least two distinct populations in flow cytometry. We used single-cell RNA sequencing to determine the transcriptional differences in peripherally and thymically induced Tregs. We sorted all living CD4+ T cells (CD45+, CD3+, CD4+) from small intestine lamina propria of four wildtype mice via fluorescence-activated cell sorting.流式细胞术（flow cytometry）分析表明，肠道调节性T细胞至少存在两种不同的亚群。本研究借助单细胞RNA测序（single-cell RNA sequencing）技术，解析外周诱导型与胸腺诱导型调节性T细胞间的转录组差异。通过荧光激活细胞分选（fluorescence-activated cell sorting），本研究从4只野生型小鼠的小肠固有层中分选获得全部活态CD4+ T细胞（CD45+、CD3+、CD4+）。
