To understand the global gene expression changes in Itga3-deficient Th17 cells, we performed RNA-seq with fate-mapped Th17 cells isolated from inguinal lymph nodes of EAE-induced mice at day 7 post EAE induction. Overall design: To acquire samples from the same host, naïve 2D2 CD4+ T cells isolated from Il17aCre/WT R26Zsgreen/WT (WTIl17a R26ZG) (CD45.1+) or Itga3fl/fl Il17aCre/WT R26Zsgreen/WT (KOIl17a R26ZG) mice were mixed at a 1:1 ratio then adoptively transferred Tcra-/- mice. On the following day, EAE was induced. ZG+ cells were FACS-sorted from iLN cells. 10 iLNs were pooled into one replicate, and three total replicates were prepared for each genotype.
