筛选某癌种中基因突变的热点位点(突变率高的位点):根据积累的NGS数据,筛选出在该癌种中突变率高的可治疗位点,用于开发PCR检测试剂盒,可以使PCR检测试剂盒更有临床应用价值。1. 数据采集:采集送检样本的肿瘤信息;
2. 基因检测:通过高通量测序技术,对样本进行多基因进行检测;
3. 数据处理:筛选出目标癌种基因突变检测结果,进一步筛选出可治疗的基因突变位点,统计各位点的突变率,在某癌种中各基因突变位点突变率=该癌种中该位点突变患者数/该癌种中进行该基因检测的患者总数,根据临床实际应用情况以及筛选出的突变位点的个数确定热点突变阈值;
4. 数据应用:以筛选出来的热点突变位点作为检测位点,开发PCR检测试剂盒。
Screening for hotspot mutation sites (i.e., sites with high mutation frequency) of gene mutations in a specific cancer type: Based on accumulated Next-Generation Sequencing (NGS) data, actionable sites with high mutation frequency in this cancer type are screened out for the development of PCR detection kits, thereby improving the clinical utility of the kits.
1. Data Collection: Collect tumor information from submitted clinical samples;
2. Genetic Testing: Perform multi-gene testing on the samples using high-throughput sequencing technology;
3. Data Processing: Screen out the gene mutation test results for the target cancer type, further select actionable gene mutation sites, and calculate the mutation frequency of each site. The mutation frequency of a gene mutation site in a specific cancer type is defined as: number of patients carrying this site mutation in this cancer type divided by the total number of patients who underwent this gene test in this cancer type. The threshold for hotspot mutations is determined based on actual clinical application scenarios and the number of screened mutation sites;
4. Data Application: Develop PCR detection kits using the screened hotspot mutation sites as the detection targets.
