Kidney organoids in vitro model enabled identification of a novel mechanism that may be used for the treatment of Fabry disease. Human inducible pluripotent stem cell line (CMC11) was obtained from the Catholic university of korea and maintained in mTesr1 medium. Total RNA was prepared from the kidney organoids and fabry kidney organoids using TRIzol (Invitrogen). Gene expression studies were performed using the Affymetrix One Cycle labeling kit. The data were summarized and normalized using the robust multi-average (RMA) method implemented in Affymetrix® Power Tools (APT). The results with gene level RMA analysis were exported and DEG analysis was performed. Statistical significance of the expression data was determined using LPE test and fold change in which the null hypothesis was that no difference exists among groups.
