Microalgal-bacterial interactions strongly influence algal physiology, yet the mechanisms shaping these associations under defined nutrient limitation remain unclear. This study aimed to determine how nitrogen, phosphorus, and vitamin B12 limitation affect the transcriptional responses of P. parvum and a defined 15-member bacterial synthetic community. Samples were obtained from controlled laboratory cultures of P. parvum grown alone in a B12-limited media, or with the SynCom under three nutrient regimes: N-limited, P-limited, and B12-limited. Biological triplicates were harvested at the end of the experiment, centrifuged, flash-frozen in liquid nitrogen, and stored at -70 C before RNA processing. Total RNA was extracted using standard phenol-based protocols followed by DNase treatment. RNA quality was verified by Bioanalyzer, and high-quality samples were used for rRNA depletion and Illumina paired-end library preparation.
