CAR T cell therapy faces obstacles in treating solid tumors. Immunomodulatory drugs especially lenalidomide has shown the capability to improve CAR T cell anti-tumor function in solid tumor therapy. However the multiple targets and drug resisdance of lenalidomide may hinder its application in combinatory therapy. IKZF3 is one of the prominent targets downstream of lenalidomide, which may be practical to improve CAR T cell function. In this study, we address that IKZF3 deficiency significantly enhances the CAR T anti-tumor function.
