Chimeric antigen receptor (CAR) T-cells have demonstrated promising efficacy, particularly inhematologic malignancies. A challenge to CAR T-cells in solid tumors is the immunosuppressivetumor microenvironment (TME), characterized by high-levels of the inhibitory cytokine, TGFB.We report results from a first-in-human phase 1 trial of prostate-specific membrane antigendirected CAR T-cells armored with a dominant-negative TGFB receptor(ClinicalTrials.gov NCT03089203).
