Effect of miR-181 and miR-1249 injection on adipose tissue transcriptomics

In this study we designed miRNAs mimetics and treated mice using in vivo jetPEI transfection reagent (Polyplus Transfection, Illkirch, France) according to the manufacturer instructions. Mice were divided randomly into three groups (n = 8/group): normal controls treated with placebo-base control, N treated with 8 ug/g miR-181b mimetic, and N treated with 8 ug/g miR-1249 mimetic. All mice were subjected to six intraperitoneal (i.p.) injections 48 hours apart between each mimetic or negative control injection. The 16h fasted mice were euthanized, and tissue were collected 48 h after the last injection. On the morning during tissue collection we measured fasted glucose level and our analysis indicated that both miR-181b and miR-1249 significantly reduced glucose level in treated animals when comparing with negative control mice (P<0.03 and P<0.0002, respectively). Additionally, from treated animals we randomaly selected n=4-6 of RNA from each group for transcriptomic analysis. Overall, we observed that miR-181 down-regulated 831 genes (FC<0.5;FDR<0.05) and up-regulated 940 genes (FC>2; FDR<0.05), while miR-1249 down-regulated 396 genes and up-regulated 875 genes (Figure 4). In Figure 4 we can observed that Pi3k/Akt/FoXO and mTOR pathways were significant regulated in both treated groups when comparing with controls.
