资源简介:
Introduction: Trypanosoma cruzi, the parasite that causes Chagas disease, requires of essential metabolites and cofactors to guarantee its survival. This parasite must supply its quota of metal ions such as copper (Cu), iron (Fe), calcium (Ca), etc. T. cruzi depends on a mitochondrial respiratory chain with the contribution of a cytochrome c oxidase type aa3 (COX) enzyme as the main oxidase. The COX enzyme, in addition to the 2 heme A molecules (an and a3), also requires Cu ions for its activity. Mitochondrial function and COX activity are essential for parasite proliferation and infectivity. This dataset contains the following files: 2022-02-02_Growth curves 1_ Starved cells plus Cu 50 a 250.xlsx (see METHODOLOGICAL INFORMATION section A) 2022-02-11_Growth curves 2_Copper trearment.xlsx (see METHODOLOGICAL INFORMATION section A) 2022-09-09_Growth curves 3_BCS and NeoCup treatment.xlsx (see METHODOLOGICAL INFORMATION section A) 2023-06_ICP.xlsx (see METHODOLOGICAL INFORMATION section B) 2024-04 to 06_ICP samples_Epis with Neo (see METHODOLOGICAL INFORMATION section B) 2022-01-17_Effect of Cu and chelators in the infection_3rd.xlsx (see METHODOLOGICAL INFORMATION section C) The compressed file “2022-11-08_Intracelular heme.7z” has 23 files (see METHODOLOGICAL INFORMATION section D) The compressed file “2024-06-20_08_Intracelular heme_Cu 500uM treatment.7z” has 19 files (see METHODOLOGICAL INFORMATION section D) 2022-08-25_Effect of Cu and chelators on metaciclogenesis.xlsx (see METHODOLOGICAL INFORMATION section E) 2022-10-17_Oxygen consumption with BCS_day 5.xlsx (see METHODOLOGICAL INFORMATION section F2) 2022-09-30_Oxygen consumption with Cu_BCS_NeoCup_day3.xlsx (see METHODOLOGICAL INFORMATION section F1) 2022-11-08_ROS test_in triplicates_3rd.xlsx (see METHODOLOGICAL INFORMATION section G) 2021-12-30al 1-3_Viability test_Cu and NeoCup.xlsx (see METHODOLOGICAL INFORMATION section H1) 2021-12-23_27_Viability test_BCS.xlsx (see METHODOLOGICAL INFORMATION section H2) 2024-06-11_Viability test on epimastigotes (see METHODOLOGICAL INFORMATION section J) The compressed file “qPCRs runned in OPUS96.7z” has 10 files (see METHODOLOGICAL INFORMATION section I): 2023-05-12 Amas Ubiquitina.csv, 2023-05-10 repetitions Amas.csv, 2023-05-05 repetitions.csv, 2022.12.07 Ubiquitin complete.csv, 2022.12.07 Ubiquitin 1.csv, 2022.11.03 FR18.csv, 2022.11.02 FR18.csv, 2022.10.13 Fet3.csv, 2022.10.12 IT.csv, 2022.10.06 CuATPase.csv METHODOLOGICAL INFORMATION Complete methodological information is available in the Readme.docx file and in the descriptive metadata of each file. Value of the data: This data is the first work that provides information on the copper metabolism in T. cruzi. Copper is an essential cofactor for aerobic organisms and at the same time is a toxic element in high quantities. For these reasons, there must be transport for this ion and at the same time it must be a controlled transport. In this work we analyzed the tolerance to Cu stress, in excess and deficiency, and in the different stages of the life cycle. The transcriptional response of a selection of genes that, we propose, are involved in the control of Cu ion homeostasis was studied.
