Although prime editing can introduce any small-sized genetic change, the efficiency of prime editor 2 (PE2) is often insufficient, limiting its applications. Here we generated a variant of PE2, named hyPE2, by adding the Rad51 DNA-binding domain between the Cas9 nickase and reverse transcriptase domains. Editing efficiencies of hyPE2 were up to an average of 2.5-fold higher than those of PE2 in cultured human cells, potentially facilitating prime editing applications
