We analyzed the effect of diet-induced obesity on liver gene expression in wild-type mice and in mice lacking p110alpha in hepatocytes. Samples were collected from mice either fed or fasted. Overall design: Mice lacking p110 alpha in hepatocytes were created by mating the floxed-p110α mouse strain (LWT) with C57BL/6J albumin-Cre transgenic mice to obtain albumin-Cre+/−p110αflox/flox mice, i.e., p110αhep-/- mice (LKO). Twelve-week-old p110αhep+/+ (LWT) and p110αhep-/- (LKO) mice were fed a HFD with 60% calories from fat (D12492, Research Diet) or a control (“CTRL”) diet (D12450J, Research Diet) during 12 weeks. All mice had free access to food and water. At the end of experiment, mice were fasted at Zeitgeber Time (ZT) 16 (with ZT0 being when the light is turned on and ZT12 when light is turned off) for 24h or fed ad libitum prior sacrifice. Mice were killed at ZT16.
