Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are to compare epidermal keratinocytes transcriptome profiling (RNA-seq) between wild type and Osmr-/- mice Methods: Epidermal keratinocytes mRNA profiles of wild-type (WT) and Oncostatin M receptor (Osmr-/-) mice were generated by deep sequencing, in triplicate. The sequence reads that passed quality filters were analyzed at the transcript isoform level with two methods: Burrows-Wheeler Aligner (BWA) followed by ANOVA (ANOVA) and TopHat followed by Cufflinks. 研究背景与目的:下一代测序(Next-generation sequencing, NGS)已彻底革新了基于系统生物学的细胞通路分析范式。本研究旨在对比野生型与抑瘤素M受体基因敲除(Osmr-/-)小鼠的表皮角质形成细胞转录组谱(RNA-seq)。实验方法:本研究通过深度测序构建野生型(WT)及抑瘤素M受体(Oncostatin M receptor, Osmr-/-)小鼠的表皮角质形成细胞mRNA表达谱,每组设置三次生物学重复。对通过质量过滤的测序读段,分别采用两种分析流程在转录本异构体水平开展解析:其一为Burrows-Wheeler比对工具(Burrows-Wheeler Aligner, BWA)结合方差分析(ANOVA),其二为TopHat与Cufflinks联用分析。
