Recent advances in DNA sequencing allow researchers to map changes in DNA methylation genome-wide in cancer and disease. However, we lack simple and robust experimental tools to site-specifically manipulate DNA methylation to discern the function of these changes. We developed a CRISPR/Cas9 DNA methyltransferase 3A (DNMT3A) fusion to induce methylation at specific loci in the genome. To test this approach we measured methylation levels using deep bisulfite sequencing of targeted loci. This CRISPR/Cas9-DNMT3A fusion approach robustly methylates DNA and can be easily reprogrammed to understand the functional consequences of methylation changes in cancer and disease.
