DNA from T. brucei Lister 427 isolate, PN221 (Figueiredo et al., PLoS Biol., 2008), was sequenced with SMRT sequencing technology (PacBio) to ~100X coverage. The reads were assembled into contigs following the RS_HGAP_Assembly.3 workflow from SMRT Analysis v2.3.0, with default parameters. Contigs belonging to megabase chromosomes were then scaffolded based on genome-wide chromosome conformation capture (Hi-C) data. For more information, see Muller et al., Nature, 2018. Then, short-read error-corrected PacBio reads and Hi-C data was used to error-correct and generate a fully-phased genome assembly. For more information, see Cosentino et al. NARGB, 2021.This BioProject contains the assembly encompassing unscaffolded central 'core' of the eleven megabase chromosomes from haplotype "A", the alelle-specific subtelomeric regions from both haplotypes and the unassembled contigs, including minichromosomes and the kinetoplastid maxicircle.
