The effects of scar resection in chronic complete spinal cord injury(SCI) and the effects of scar resection combined with decellularized hydrogel (scaffold)

This study examines the molecular effects of surgical scar resection in complete chronic spinal cord injury (SCI) through RNA sequencing. Results show that scar resection alters gene expression, with downregulation of axonal growth inhibitors and upregulation of neuroprotective genes. Additionally, the application of a decellularized extracellular matrix (dECM) hydrogel as a scaffold promoted angiogenesis. Gene Ontology analysis indicated enrichment of immune-related processes, suggesting that scar resection improves the spinal cord microenvironment for neural repair. These findings indicate the potential of combining scar resection with scaffolding to support recovery after chronic complete SCI.
本研究通过RNA测序(RNA sequencing)技术,探究了慢性完全性脊髓损伤(spinal cord injury, SCI)患者接受手术瘢痕切除术后的分子生物学效应。结果显示,瘢痕切除术可改变基因表达谱:轴突生长抑制因子的表达显著下调,而神经保护基因的表达则显著上调。此外,采用脱细胞细胞外基质(decellularized extracellular matrix, dECM)水凝胶作为支架,可促进血管生成。基因本体论(Gene Ontology, GO)分析表明,免疫相关生物学过程出现富集,提示瘢痕切除术可改善神经修复所需的脊髓微环境。上述研究结果表明,将瘢痕切除术与支架干预相结合,有望助力慢性完全性脊髓损伤后的功能恢复。
