HSC subtypes with distintic lineage restrictions have been previously reported. Here we examine the chromatin landscapes of HSC subtypes with P-restricted, PEM-restricted or multilineage in vivo output. Single cell transplantations of Vwf+ HSCs from mouse bone marrow were performed to generated primary recipients showing lineage-restricted reconstitution over 16 weeks : P-restricted, PEM-restricted and multilineage. Donor-derived Gata1- HSCs were isolated from recipients' bone marrow for bulk ATAC-seq (500 cells per replicate, 3 replicates for PLT-HSC, 6 replicates for PEM-HSC, 4 replicates for MUL-HSC).
